When your CrossFit friends tell you about their W.O.D and now you can’t move... 🥵🏋️‍♀️💪🏻 I like to keep my life relatively drama free, but in biology there are 2 proteins that are constantly hot and cold. You guessed it! Actin and Myosin!!! Muscles are awesome because they take chemical potential energy and turn it into mechanical energy thanks to the constant coupling and separation of Actin and Myosin. Since muscles are essentially effective in thanks to those 2 proteins it’s worth remembering that proteins tend to follow 2 main rules. 1️⃣ Proteins like to change shape when stuff binds to them. 2️⃣Changing shapes can allow proteins to bind or inhibit their binding to other stuff. At rest Actin and Myosin don’t touch and have chaperones of Tropomyosin and Troponin blocking their interaction. Those guys can be bought off for the low low price of ATP and calcium. Happy Friday friends. This week has been a busy week working on the microscopes. Been gathering lots of images for quantitative analysis of actin in plant roots. Actin (green filaments in the image) is a major component of the cytoskeleton (Cell skeleton). In addition to actin filaments (aka microfilaments), the cytoskeleton consists of microtubules and intermediate filaments. These microscopic structures, specify cell shape, facilitate movement of cell components or in some cases, the cell itself, and offer structural support to the cell. . Our laboratory 🔬 studies how the cytoskeleton specifies root system architecture in model and crop plants 🌱 on Earth 🌍 and in the microgravity (reduced gravity) environment of Spaceflight. More on the latter project in a future post. . The long term, practical goal of the lab is to contribute to the development of plant cultivars that are more resilient to the harsh environments of the USA Southern plains (e.g. drought, low nutrients) and beyond, including future farms and space colonies on the moon 🌚 and Mars. . These are so called N1E-115 cells, This immortalized cell-line was isolated in 1971 from a mouse neuroblastoma. Using phalloidin, a toxin containing a fluorescent tag that binds to filamentous actin I was able to visualize the actin cytoskeleton. The bright spots are spots are called invadosomes, these are actin-rich adhesion structure. These remarkable images were created by Dr. Rashi Karunasinghe during her IBRO-USCRC North American Lab Activity where she was hosted by Dr. Hari Shroff at the National Institute of Biomedical Imaging and Bioengineering in Bethesda, MD, USA, for a month. . . The video is a recording of actin particles moving through developing neurons. I went to Cornell for undergrad (Go Big Red!) where I first got involved in research studying cardiovascular disease and development. I started college with plans to go to medical school, but after spending so much time doing research, I couldn't imagine a career without it. It's an area where I can flex my curiosity and constantly try new things. In the end, I decided to apply to MD/PhD programs where I could potentially bring some of my science into the clinic to help patients. After graduating, I spent a year at the National Institutes of Health studying the role the myosin in development, specifically in the heart and testes. Here are Penn, my first rotation was spent studying lymphatic vessel development and regulation of lymphatic endothelial cell permeability. This is why you guys see so many images of the heart, testes, lymphatic endothelial cells, and cytoskeleton. I'm hoping to do my PhD development and cell biology. Penn is an incredible place to do research, and just this past year, we saw the first gene therapy for a genetic disease and CAR-T cell immunotherapy for cancer become FDA approved. I'll be attending the ASCB/EMBO meeting in San Diego this weekend and super excited to see the incredible research (and microscopy!) being done by fellow cell biologists.
